Biochemistry ยท Spring 2026
Biochemistry Lab Capstone
Investigated the effects of point mutations on the structure, stability, and enzymatic activity of ฮฒ-glucosidase B (BglB), a TIM-barrel enzyme involved in carbohydrate metabolism. Generated mutant constructs via site-directed mutagenesis, expressed recombinant protein in E. coli, and purified using Ni-NTA affinity chromatography. Characterized enzymatic function using pNPG spectrophotometric assays, SDS-PAGE, and thermal stability analysis. Compared wild-type and W123Y mutant variants to assess how a single amino acid substitution alters protein stability and catalytic activity.
